Thermo Scientific QuEChERS standard step-by-step workflow
As shown in the workflow diagram below, the QuEChERS method comprises four basic steps:
- Homogenize Sample (Fruit or Vegetable): Begin by thoroughly homogenizing the sample, such as a fruit or vegetable, to ensure a uniform consistency. This step is crucial for accurate and representative analysis, as it ensures that the analytes are evenly distributed throughout the sample.
- Extract Analytes of Interest: Extract the analytes of interest from the homogenized sample using specific extraction salts. These analytes can include pesticide residues, veterinary drugs, and/or mycotoxins. The extraction process involves adding the salts to the sample, which facilitates the release of the target compounds into the solution.
- Clean-Up the Resulting Supernatant: Clean up the resulting supernatant using dispersive solid phase extraction (dSPE). This step involves adding sorbents to the supernatant to remove unwanted matrix components that could interfere with the analysis. The dSPE process helps to purify the extract, ensuring that the final sample is free from contaminants and ready for accurate analysis.
- Analyze by GC-MS or LC-MS: Finally, analyze the cleaned-up extract using gas chromatography-mass spectrometry (GC-MS) or liquid chromatography-mass spectrometry (LC-MS). QuEChERS extraction and clean-up kits for AOAC .01 method
The AOAC .01 QuEChERS Method, developed by Steven J. Lehotay and his co-workers, refines the original method by incorporating higher concentrations of acetate buffering for enhanced buffering strength. Buffering is essential in the QuEChERS procedure as it stabilizes pH during extraction and clean-up steps, ensuring accurate analysis of pH-dependent pesticides and other residues.
